Specific Aims This project involves investigation of marijuana cue-reactivity as a bio-assay for medication development in adolescents with marijuana use disorders. Marijuana use disorder continues to be a major problem among adolescents in the U.S. Despite this, there is little work examining medication treatment for marijuana abuse and dependence in this population. As has been shown with other drugs of abuse, craving is likely involved in maintenance of marijuana use, and may be crucial in relapse during abstinence. Hence, a medication that can successfully reduce craving may prove to be useful in the treatment of those seeking to achieve sustained abstinence from marijuana use. There are no published reports of marijuana cue-reactivity in adolescents or randomized double blind medication trials for marijuana use disorders in adolescents. Large clinical trials require significant resources and are difficult to conduct, especially in adolescents. Hence, we propose an exploratory/developmental grant to test whether buspirone will potentially dampen craving in adolescents with marijuana use disorders (abuse/dependence) in a cue-reactivity bioassay paradigm developed to screen medications for marijuana use disorders. The primary specific aim of the proposed project is: 1) To explore potential efficacy of buspirone in reducing marijuana craving among adolescents with marijuana use disorder. The secondary specific aim of the proposed project is: 2) To explore cue-reactivity as a bio-assay to screen medications for treatment of marijuana use disorders in adolescents.
