In order to clarify the mechanism of cause and development of osteoarthritis, the relation between mechanical stress and cytokines which play an important role for osteoporosis was investigated using a chondrocyte-like cell line established in our department.As the results, while it was evidenced that centrifugal force at 2000 rpm until 60 minutes accelated cell proliferation with time-dependency, excess force (3000rpm) induced cell death. 80 % of cells for 60 min.at 3000 rpm were dead. We already indicated that appoptosis of cells was induced by TNF-alpha considered as one of critical cytokine for post-osteomenoposal osteoporosis. Therefore the effects of TNF-alpha under mechanical stress was studied. It was showed that TNF-alpha was able to stimulate cell death under suitable mechanical stress (2000 rpm for 60 min.). These results indicated that the cells was not only injured by excess mechanical stress, but also injured by suitable stress, if TNF-alpha was containing there.
