Administrative supplement application for equipment purchase
This is an administrative supplement application for the purchase of a AKTA_Pure Fast Pressure Liquid
Chromatography (FPLC) system from Cytiva. The AKTA_Pure FPLC is a medium flow, medium pressure
chromatography system that allows the semi-automated, rapid, and high-resolution chromatographic
separation of proteins by ion-exchange, hydrophobic interaction, gel-filtration, and affinity chromatography.
Protein purification and analysis, enabled by the FPLC, is a central and indispensable requirement for all the
projects being pursued in my laboratory as part of my NIGMS-funded R35/MIRA award. The new AKTA_Pure
FPLC being requested in this supplement will directly replace a malfunctioning AKTA_Explorer instrument that
I purchased 13 years ago (in 2009) when I first started my laboratory at Stanford and that is no longer
serviceable. We provide documentation from the current vendor (Cytiva) that support for this old
AKTA_Explorer system lapsed in 2018. Parts are not available, vendor support is not available and the
operating system required to run instrument software is not supported by either Microsoft or Cytiva. Our
intensive use (~20-30 hours per week) and project-specific configurations preclude the cost-effective and
time-efficient use of chromatography systems in core facilities at Stanford. Finally, the improved automation
and software that comes with the new system will dramatically accelerate our workflows, improve
reproducibility and provide up-to-date training in the core skills of protein purification and analysis for trainees
working on our NIGMS-funded projects. The Biochemistry Department at Stanford will support this purchase
with both funds for instrument service and space to house the AKTA_Pure FPLC system.
