LncRNA is capable to regulate target gene expression by competing against mRNA in binding with miRNAs, which is termed as the competitive endogenous RNA (ceRNA) mechanism and has become a hot topic of studies in immunity. To date, lncRNA-mediated ceRNA has been poorly investigated in fish. Sarm is a recently discovered TLR adaptor and exerts crucial effects on immune defense, but the mechanism of which remains to be deciphered. In our previous study, we have found that lncRNA78 was the key immune regulatory factor in the lncRNA-mRNA functional enrichment network of Japanese flounder in response to Vibrio anguillarum infection, and the downregulated expression profiles of lncRNA78 and sarm after V. anguillarum challenge exhibited significantly positive correlation, which is the typical characteristic of ceRNA. Based on these observations, in this proposal, we will construct a ceRNA network of Japanese flounder in response to V. anguillarum infection, identify the target miRNA(s) of lncRNA78, and uncover the ceRNA mechanism of lncRNA78-mediated regulation of sarm, as well as the role of lncRNA78-sarm in the immune response to bacterial infection. The results of our study will pave the way for understanding the mechanism in regulating sarm, and extend the contents of study on ceRNA immune regulatory mechanism in fish.
