Objective: To establish an HPLC method for simultaneously determining the contents of malonyl ginsenosides and neutral ginsenosides in American ginseng. Methods: The analysis of the 80% methanol extract of American ginseng was carried out on a COSMOSIL 5C₁₈ - MS - Ⅱ column (250 mm × 4.6 mm, 5 μm); the mobile phase was acetonitrile - phosphate buffer, with gradient elution; the volume flow rate was 1.0 mL/min; the column temperature was 25 °C, and the detection wavelength was 203 nm. Results: The malonyl ginsenosides and neutral ginsenosides had good linear relationships in the range of 0.010 - 0.640 mg/mL (r > 0.9991), the average sample addition recoveries were 96.2% - 104.6%, and the RSDs were 0.74% - 2.58%. Conclusion: This method is simple and has good reproducibility, and can be used for the quality control of American ginseng.
目的建立HPLC法同时测定西洋参中丙二酰基人参皂苷、中性人参皂苷的含有量。方法西洋参80%甲醇提取液的分析采用COSMOSIL 5C_(18)-MS-Ⅱ柱(250 mm×4.6 mm,5 μm);流动相乙腈-磷酸缓冲盐,梯度洗脱;体积流量1.0 mL/min;柱温25 ℃,检测波长203 nm。结果丙二酰基人参皂苷、中性人参皂苷在0.010~ 0.640 mg /mL范围内线性关系良好(r>0.999 1),平均加样回收率96.2%~ 104.6%,RSD 0.74%~ 2.58%。结论该方法简便、重复性好,可用于西洋参的质量控制。
