Taking the co-culture system of primary sheep trophoblast cells (STCs) and endometrial luminal epithelial cells (ELECs) as the research object, by adding different concentrations of 17β-estrodiol (E₂) and progesterone (P₄) to study their effects on the formation of multinucleated cells in the sheep chorionic trophoblast, providing an experimental basis for exploring the growth and development of the sheep placenta. Firstly, the Giemsa staining method was used to observe the number of multinucleated cells in the chorionic trophoblast to determine the optimal working concentrations of E₂ and P₄; then, according to the optimal working concentrations, a combined use group of E₂ and P₄ was established to observe its effect on the number of multinucleated cells. The results showed that the addition of E₂ alone significantly increased the number of multinucleated cells in the trophoblast, and the addition of P₄ alone significantly decreased the number of multinucleated cells in the trophoblast, and with the increase in the concentrations of E₂ and P₄, the number of multinucleated cells in the trophoblast all showed a trend of first increasing and then decreasing, and when the concentrations of E₂ and P₄ were both 10⁻⁷ mol/L, the number of multinucleated cells in the trophoblast was the largest and showed a significant difference compared with the control group (P < 0.05); after the combined use of E₂ and P₄, the promoting effect of E₂ and the inhibitory effect of P₄ were also shown. The results indicate that in the middle stage of pregnancy, the addition of E₂ has a promoting effect on the formation of multinucleated cells in the chorionic trophoblast, while the addition of P₄ has an inhibitory effect on its formation, which lays a foundation for the research on the mechanism of sheep embryo development.
以绵羊绒毛膜滋养层细胞(primary sheep trophoblast cells,STCs)和子宫内膜腔上皮细胞(endometrial luminal epithelial cells,ELECs)的共培养体系为研究对象,通过添加不同浓度的雌二醇(17β-estrodiol,E_2)和孕酮(progesterone, P_4)以研究其对绵羊绒毛膜滋养层多核细胞形成的影响,为探究绵羊的胎盘生长发育提供试验依据。首先采用姬姆萨染色法观察绒毛膜滋养层多核细胞的数量,以此来确定E_2和P_4的最佳工作浓度;而后根据最佳的工作浓度设立E_2和P_4联合使用组,来观察其对多核细胞数量的影响。结果显示:单独添加E_2使滋养层多核细胞数量明显增多,单独加入P_4使滋养层多核细胞的数量明显减少,且随着E_2和P_4浓度的升高滋养层多核细胞的数量均表现出先增多后减少的趋势,而当E_2和P_4浓度均为10~(-7) mol/L时,滋养层多核细胞的数量最多,且与对照组相比呈显著性差异(P<0.05);E_2和P_4联合使用后,同样表现出E_2的促进作用和P_4的抑制作用。结果表明,在妊娠中期,E_2的加入有促进绒毛膜滋养层多核细胞的形成作用,而P_4的加入具有抑制其形成的作用,这为绵羊胚胎发育机制的研究奠定基础。
