The determination of targets is the core of clarifying the mechanism of action of traditional Chinese medicine and the key to constructing the correlation between chemical components and pharmacological effects. Given the diversity of traditional Chinese medicine and the complexity of chemical components, traditional target screening methods have limitations, and target discovery has become a bottleneck in the research on the mechanism of traditional Chinese medicine. Label-free small molecule probe technology is an emerging technology in the field of chemical proteomics. Its prominent features are that it does not require structural modification and transformation of small molecule components and is generally applicable to complex chemical components of traditional Chinese medicine. At the same time, this method screens targets based on proteomics, which is conducive to the discovery of new targets and new pathways of the action of traditional Chinese medicine. Given the application potential of this method in the research of traditional Chinese medicine, this article reviews its analytical principles and application status, summarizes its general analytical steps, in order to promote the application of this method in the elucidation of targets of chemical components of traditional Chinese medicine.
靶点的确定是阐明中药作用机制的核心,是构建化学成分与药效作用相关性的关键。鉴于中药的多样性、化学成分的复杂性,传统的靶点筛选方法有所局限,靶点发现已成为中药机制研究的瓶颈。无标记小分子探针技术是化学蛋白组学领域的新兴技术,其突出特点为无需对小分子成分进行结构修饰与改造,普适于复杂的中药化学成分,同时该方法基于蛋白质组进行靶点筛选,利于发现中药作用的新靶点、新途径。鉴于该方法在中药研究中的应用潜力,该文综述了其分析原理与应用现状,总结了其一般性分析步骤,以促进该方法在中药化学成分靶点阐明中的应用。
