By analyzing and comparing the total number of chicken red blood cells (RBC), hemoglobin content (HGB), and packed cell volume (PCV) measured by traditional methods and a blood cell analyzer, it was explored whether a blood cell analyzer could be used to replace traditional methods for collecting relevant blood data of broilers. 120 AA commercial broiler chickens were reared conventionally and randomly divided into two groups at 15 days of age. The control group was reared at normal temperature (22 - 24 °C); the low - temperature group was gradually cooled at a rate of 1 - 2 °C per day starting from 15 days of age, and by 21 days of age, the indoor temperature was maintained at 10 - 12 °C until the end of the experiment. At 28 and 42 days of age, 12 broiler chickens were randomly selected from each group and venous blood was collected. RBC, HGB, and PCV were measured by traditional methods and an analyzer respectively. The results showed that the RBC and HGB measured by the instrument could not reflect the actual values of broiler blood cells, and the PCV was close to the actual value; variance analysis was performed on the data measured by the two methods respectively, and the results showed that the changes in differences between groups were similar, and the detection results of the two methods were extremely significantly correlated (P < 0.01). The research results indicate that when analyzing the differences between groups of RBC, HGB, and PCV values of broiler blood measured by a fully automatic blood cell analyzer, it has reference significance.
通过分析比较传统方法和血细胞分析仪测定的鸡红细胞总数(RBC)、血红蛋白含量(HGB)和红细胞压积(PCV),探讨能否应用血细胞分析仪代替传统方法采集肉鸡相关血液数据。120只AA商品代肉仔鸡常规饲养,15日龄时随机分成两组。对照组常温(22~24 ℃)饲养;低温组从15日龄开始以每日1~2 ℃的速度逐渐降温,到21日龄时,室内温度维持在10~12 ℃直到试验结束。在28和42日龄时,每组随机抽取12只肉鸡,静脉采血。分别用传统方法和分析仪分别测定RBC、HGB和PCV。结果显示:仪器测定的RBC、HGB不能反映肉鸡血细胞的实际数值,PCV与实际值接近;分别对两种方法测得数据进行方差分析,结果显示组间差异变化相似,两种方法检测结果极显著相关(P<0.01)。研究结果表明,全自动血细胞分析仪测定的肉鸡血液RBC、HGB和PCV值进行组间差异分析时具有借鉴意义。
